Cancer Angiogenesis and Tumorigenesis Regulation: Molecular Mechanisms and Genetic Predisposition
Cancer is an incredibly challenging and devastating condition. While there has been significant progress in the field of cancer therapeutics, the emergence of drug resistance has posed a formidable obstacle to achieving widespread success in treatment.
Our investigative pursuits revolve around the identification of Tumor Suppressor Genes (TSGs) and Oncogenes having roles in multiple hallmarks of cancers. Currently, our efforts are bifurcated into two distinct yet synergistic streams. One faction is dedicated to validating and comprehending the molecular mechanisms of potential oncogenes and TSGs with dual roles in tumorigenesis, while the other is focused on discerning the genetic and molecular intricacies of gallbladder cancer.
We are pioneers in revealing the dual role of FRG1. Initially known for its connection to fascioscapulohumeral muscular dystrophy, our research unveiled a decrease in FRG1 expression in tumor tissues of the stomach, colon, and oral cavity. We revealed its significant involvement in regulating prostate cancer traits in a paracrine manner. Recognizing the widespread threat of breast cancer metastasis, we embarked on a journey to identify FRG1’s role, which led to the revelation that reduced FRG1 expression facilitates breast cancer progression via the GM-CSF/MEK-ERK axis. Simultaneously, FRG1's role in tumor angiogenesis emerged, as we discovered its promotion of angiogenic properties in HUVECs, regardless of breast cancer subtypes. This revelation was driven by the depletion of FRG1, which activated the FGF2-mediated signaling cascade, subsequently stimulating the angiogenic pathway in HUVECs. Venturing deeper into the molecular intricacies, we figured out FRG1's pervasive role in the regulation of the Nonsense-mediated mRNA Decay pathway genes and its interaction with EEF1A and IQGAPs.
EEF1A2 is a translation-elongation factor that is expressed only in specialized tissues. Oncogenic switch-on of EEF1A2 has been reported in multiple types of cancers. Notably, our studies illuminated its positive association with ER receptor in MCF7 cells, while inducing a robust metastatic program via ERK pathway activation in MDA-MB-231 (triple-negative cell line). This pro-tumorigenic role extended to brain tumors, where elevated EEF1A2 levels correlated with PI3K, underscoring its broad impact. We have unravelled that in breast cancer EEF1A2 and HIF1A regulate each other in a positive feedback loop, which has crucial implications in drug development. Our investigations shed light on the intricate interplay of IQGAPs, exposing IQGAP2's inhibitory effect on the oncogenic activity of IQGAP1. This mechanistic insight holds promise for therapeutic exploitation. We plan to identify the significance of FRG1, EEF1A2, and IQGAP interaction on molecular signaling and its implications in tumorigenesis.
We have a special interest in gallbladder cancer, a disease disproportionately prevalent in specific regions, including Odisha; we are driven by an imperative need for early detection and targeted chemotherapy. So far, we have identified various genetic risk factors. Our pioneering work on MMP promoter variants uncovered their role as risk factors in GBC pathogenesis. These variants, particularly rs1004030 and rs1003349, housed in critical cis-regulatory elements, demonstrated allele-specific binding with MYB and SOX10, dictating MMP14 expression. This newfound understanding unveils a novel mechanistic role in GBC pathogenesis. Simultaneously, our investigation into bile acid intermediates provides a holistic perspective on their proliferative effects and genetic predisposition.
In summation, our research endeavors are rooted in a resolute commitment to comprehending the pivotal roles of candidate genes, unveiling the intricate cellular and molecular pathways governing tumor progression. This knowledge forms the bedrock for formulating innovative therapeutic strategies in the battle against cancer.
